Preliminary Topic: Development of novel CRISPR-based genome editing tools to edit DNA and RNA genomes
Short Description of the Internship Work: Current CRISPR-based genome editing tools rely on traditional homologous recombination followed by counterselection methods (limited due to its toxicity), on base editing through excision or deamination of DNA bases (limited to single base substitution) or, on prime editing technologies (limited to eukaryotes and to small edits).
In our previous work, we introduced a new field in genome engineering which is termed append editing (available online soon). Append Editing (AE) is unique as it installs bulky chemical moieties on DNA, blocking DNA replication and mediating in turn homologous recombination. In this project, we will develop further the AE technology by working with novel proteins and novel mechanisms to engineer dsDNA, ssDNA, dsRNA or ssRNA genomes.
The outcome of this project will expand the toolbox for genome engineering and will give access to myriads of potential industrial and clinical applications
In this internship you will conduct some of the following techniques:
Microbial cell culture, bacteriophage propagation, cloning, genome editing, microscopy, cytometry, cell-free transcription-translation (TXTL), Next Generation Sequencing (NGS).
