Preliminary Topic: Development of a DNA-, RNA-, protein-, and transformation-free genome editing tool for bacteria
Short Description of the Internship Work: Genome editing in bacteria typically involves complex and time-consuming protocols. These methods are especially challenging for non-model bacteria, making it difficult to study or modify species beyond a few well-known species like E. coli, B. subtilis, and S. aureus (amongst others).
In this project, we aim to use a novel approach for bacterial genome editing that bypasses the need for introducing and maintaining plasmid DNA in the bacterial cells. Additionally, this method does not rely on the expression of RNAs or proteins for the editing process. This technique will allow us to edit the genomes of bacteria that are typically difficult to transform, either in isolation or within complex microbial communities. The development of this method will result in broadening the range of bacteria we can engineer, will accelerate and simplify the transformation process, and will give access to myriads of potential industrial and clinical applications.
In this internship you will conduct some of the following techniques:
Microbial cell culture, cloning, genome editing, microscopy, cytometry
