Preliminary Topic: Development of Genome Editors
Short Description of the Internship Work: The use of prokaryotic CRISPR-Cas adaptive immune systems, such as Cas9, has heralded a significant advancement in genome editing. Cas9 has simplified, expedited, and enhanced the efficiency of cellular genome editing processes. Nonetheless, challenges such as the substantial size of these proteins, their limited specificity, and other undesirable activities impose constraints on the practical application of Cas9-based genome modification technology. To address these issues, ongoing efforts in the Pausch laboratory are dedicated to the development of alternative genome engineering approaches centered around existing or novel RNA-dependent CRISPR-Cas systems. Throughout the internship, the objective will be to characterize and potentially adapt these new or existing systems, making them suitable for genome engineering applications, thereby broadening the array of tools available for genome manipulation
